Hello!

We love Little Snitch, we are fans too. It's the best application firewall for OS X. :)
Murus and Little Snitch can't be compared, they are two different things.
Using a network firewall and an application firewall together can be done, but honestly I never tried it. It may be a good idea in some scenarios. Of course you will face an increased complexity in setting up your network configuration.
My educated guess is that a connection must be passed by both firewalls in order to pass. However I have no idea about how Little Snitch interacts with Murus (PF) features like port knocking, adaptive firewall and NAT.
Try it :) And let me know what happens.

All good pf and Little Snitch, filtering flow should is
inbound: pf -> little snitch
outbound: little snitch -> pf
